Abstract:Abstract: Two transcriptional activation systems developed from CRISPR/Cas9, namely Cas9-p300 and dCas9-VPR, were employed to activate germ cell specific genes, and the differences between them in activation efficiency of target genes were compared. Real-time quantitative PCR results showed these two activation systems can activate the expression of most target genes. Cas9-p300 system activated the BLIMP1 gene more efficiently, while dCas9-VPR system activated NANOS2, DAZL and SOX17 genes more efficiently. At the same time, there was no significant difference between these two systems in activation efficiency of DDX4 and TFAP2C genes. Therefore, the study revealed that the different transcriptional activation systems have different efficiencies in activating germ cell specific genes. This will provide a theoretical basis for study of germ cell specialization by transcription activation system, and shed lights on cell fate determination in other developmental systems.
