Abstract: Abstract: Aptamers are single-stranded RNA or DNA sequences which are selected in vitro from a synthetic random oligonucleotide library using the systematic evolution of ligands by exponential enrichment (SELEX) technology. They can be folded into a specific three-dimensional spatial structure and bind to the target substance with high affinity and specificity. In recent years, nucleic acid aptamers obtained by whole-cell SELEX have great application potential in the disease-related field, especially in recognition of cellular molecules and discovery of biomarkers. However, there are still many problems constraining the development of nucleic acid aptamers. The process of screening whole cells is complicated and difficult, and the selected nucleic acid aptamers have poor performance. Obtaining efficiently nucleic acid aptamers is the key to their application. Herein, the advances of new methods and new strategies of cell-SELEX technology and the ap-plication of aptamers in tumor research were reviewed, which would provide reference for relevant researchers.
