Abstract:Abstract: The presence of amyloid β-protein (Aβ) on the surface of nerve cells is one of the major pathologic features of Alzheimer’s disease (AD). Amyloid precursor protein (APP), which is closely related to AD pathology, is cleaved by the β- and γ-secretases successively, causing the generation of Aβ, sAPPβ and AICD fragments. Aβ can trigger endoplasmic reticulum stress, mitochondrial dysfunction and so on, but the effects of the sAPPβ and AICD fragments on cells remain further research. In this study, SH-SY5Y cells were transfected with plasmids carrying sAPPβ, Aβ and AICD genes, respectively, mediated by lentivirus. The cell viability, cell membrane integrity, intracellular reactive oxygen species (ROS) level and mitochondrial membrane potential were assessed. The results showed that overexpression of Aβ or AICD fragments could cause decreased cell viability, damage to cell membrane integrity, increased intracellular ROS level, and depolarization of mitochondrial membrane.
