Abstract:Abstract: The association of genes in the RANK/RANKL/OPG and WNT signaling pathways with hip bone strength was investigated among the middle-aged and elderly males. A total of 788 males aged over 40 years were included in the study. They were recruited from Guangzhou, China. Ten single nucleotide polymorphisms in 6 genes were genotyped using MassArray. Bone mineral density (BMD), cross-sectional area (CSA), average cortical thickness (ACT), section modulus (SM) and buckling ratio (BR) at the femoral neck were measured by dual-energy X-ray absorptiometry. The relationships between the genes and bone phenotypes were detected using multiple linear regression analysis. The results showed that the minor allele C of rs3134069 in OPG gene was negatively associated with BMD and ACT (β =-0.017, P=0.024 for BMD; β=-0.004, P=0.040 for ACT). In the LRP5 gene, the minor allele T of rs3736228 was related with the lower BMD, CSA, ACT (β=-0.050~-0.003, P<0.050) and higher BR (β=0.242, P=0.043), and the minor allele G of rs491347 was associated with the reduction of BMD (β=-0.012, P=0.046). This study supported that the OPG and LRP5 genes were associated with hip bone strength of the middle-aged and elderly males in Guangzhou.
