Abstract:Abstract: O-fucosylpeptide 3-beta-N-acetylglucosaminyltransferase (Lunatic Fringe, LFNG) is closely related to Notch signaling. It plays a dual role for Notch signaling in different tissues and cells. To study the exact role of LFNG expression for Notch signaling in acute leukemic B cells, the expressions of LFNG in human acute B-lymphoblastic leukemia cell line BALL-1, human B-lymphoblastoid cell line HMy2.CIR and normal B cells were analyzed by real-time PCR and Western-blot. In addition, LFNG was inhibited by siRNA in BALL-1 to explore the effect of LFNG expression on the Notch signaling. The results showed that the expression level of LFNG in BALL-1 leukemic B cells was far higher than that in HMy2.CIR and normal B cells and down-regulation of LFNG inhibited the Notch signaling in BALL-1 cells, which suggested that the abnormal expression of LFNG in leukemic B cells promotes the Notch signaling.
