Abstract:Abstract: Polyamine analogues interfere with the normal metabolism of polyamines, and deplete intracellular polyamines that are necessary for fast-growing tumors. It was performed with MTT, the flow cytometry, the agarose-gel electrophoresis, Western-blot and transwell technique to investigate the effects of polyamine analogue tetrabutyl propanediamine on proliferation, apoptosis and migration of human PC-3 prostate cancer cells. It was showed tetrabutyl propanediamine significantly inhibited proliferation and migration of PC-3 cell and the inhibitory effect was in a time- and dose-dependent manner. After tetrabutyl propanediamine treatment, DNA fragmentation of chromosomal DNA, typical for apoptotic cell, was observed and the contents of anti-apoptotic protein Bcl-2 was significantly deceased, but pro-apoptotic protein cytochrome C was significantly increased in the cytosol. In addition, tetrabutyl propanediamine also induced inhibition of cell cycle and deceased cell migration. As mentioned above, tetrabutyl propanediamine can inhibit the proliferation and migration of human prostate cancer PC-3 cells by inhibiting cell cycle and inducing apoptosis.
